In vitro study | [MK-801 inhibition of N-methyl-D-aspartate-induced [MK-801 causes continuous, persistent blockade of NMDA-induced currents in the hippocampus. MK-801 (< 500 μm) concentration-dependently prevents LPS-induced microglial activation in Mg BV-2 cells, even MK-801 prolonged administration in the presence of NMDA, this was accompanied by an increase in Cox-2 protein expression. In BV-2 cells, MK-801 (< 500 μm) induced microglial TNF-α export with an EC50 of 400 μm. |
In vivo study | Treatment of mice with MK-801 (1 mg/kg) before each METH injection reduced the extent of 55% DA depletion in the striatum of mice. In the mouse striatum, MK-801 (1 mg/kg) attenuated the effect of METH on microglia activation. In rats, cocaine-related memory reactivation in the CPP environment preceded MK-801 (0.05 mg/kg or 0.2 mg/kg, I. p.) administration attenuates subsequent cocaine-induced recovery without interference occurring in rats and without reactivation in the CPP environment. MK-801 (0.2 mg/kg, I. p.) prior to two reactivations in the cage did not inhibit subsequent cocaine-induced reconstitution. |
